Hematological analyzers, depending on the design, work on their own hematological reagents (diluent, enzyme, lytic), but there is much in common between them.

The set of hematological reagents includes:

isotonic diluent (diluent);

cleaning solution (enzymatic).

In 18-parameter analyzers, pulses are counted when thecell passes through a narrow channel or aperture between two electrodes. After entering the blood sample into the analyzer, the diluent (isotonic diluent) conducts the dilution, since when two cells are present in the channel at the same time, they are registered as one pulse. For this, an isotonic diluent (diluent) is used - a buffer solution with certain values of pH, conductivity and osmolarity, and maintains a fixed volume of cells.

The second solution of a set of hematological reagents is a lysing or lytic (hemolytic). The lysing solution destroys erythrocytes into small fragments and converts different forms of hemoglobin into a single one.

Due to the difference in the dosing schemes of samples and reagents, in Medonic analyzers, 4 ml of lithic is added to the first dilution, and in Micros-60, MicroCC-18 analyzers it is many times less.

After lysis of erythrocytes, only leukocytes remain in the sample. Subtracting from the first count of high-amplitude pulses the count of high-amplitude pulses of the second count — leukocytes, the analyzer will give the number of erythrocytes.

In addition to an isotonic diluent for diluting blood and a hemolytic agent for destroying erythrocytes, a washing solution - an enzyme, a detergent - is required for work on the analyzer. The enzyme is used only for washing the working systems of the device and has a positive effect on the stability and accuracy of the analyzer. It is necessary to use detergent solutions and enzymes for cleaning analyzer systems after each analysis.

So, for example, due to the differences in the hydraulic system washing schemes, in the Cell Dyn, PCE-210 analyzers, washing is carried out after the analysis of each sample and detergent solutions are suitable for this, while in the Advia 60 analyzers, washing is carried out after a given number of analyzes and for this, an enzyme is required - a more effective enzymatic washing reagent. In the Sysmex KX-21 analyzers, the use of washing solutions connected to the system is not provided, and periodic cleaning of the analyzer is carried out with the help of hypochlorite solutions.

Periodically, the analyzer needs calibration using standard materials, as the electronic and mechanical components of the device, sensors, and pump age over time and change their technical parameters.
